The scarcity of nonrenewable resources and the increase in environmental pollution have intensified the search for materials that exhibit specific characteristics and are nontoxic, renewable, and sustainable. Thus, the objective of this work was to produce natural polyphenol adhesives reinforced with rice husk and its ash to increase the mechanical resistance and moisture resistance of the glue line in wood bonded joints. Polyphenols were extracted from the bark of Stryphnodendron adstringens (Mart.) Coville (barbatimão). Adhesives were produced with a 50 % solid and 50 % liquid composition. Rice husk and husk ash underwent X-ray fluorescence analysis (XRF). Adhesives and reinforcement material were characterized by Fourier transform infrared (FTIR) and thermogravimetric analyses (TGA). The adhesives were glued in a mechanical press in specimens made of Pinus elliottii, which were subjected to shear testing of the wet and dry glue line. As a result, the chemical components present in rice husk and its ash positively influenced the properties of the adhesives. The mechanical glue line shear test showed that the adhesive reinforced with rice husk ash did not show a statistically significant difference. However, natural adhesives based on polyphenols from barbatimão strengthened with rice husk and ash showed improved properties, demonstrating how much it pays to use the residue of rice production to reinforce the matrix of tannin adhesives. Thus, it can be determined that reinforcement with rice husk and ash is efficient in improving some properties of natural adhesives based on polyphenols.

The 5-alpha-reductase enzyme, present in pilosebaceous units, plays a crucial role in the appearance of cutaneous hyperandrogenism manifestations (hirsutism, acne, and androgenetic alopecia). Its inhibition is an excellent strategy to reverse these conditions. Given the limitations of existing treatments, with transient effects and delayed therapeutic response, as well as the possibility of causing undesirable side effects, this study sought to develop new drug delivery systems to overcome these limitations. In other words, innovative stimuli-responsive hybrid nanoparticles were synthesized using silica/natural polysaccharides, encapsulating 5-alpha-reductase enzyme inhibitors derived from the plant Stryphnodendron adstringens (Mart.) Coville (commonly known as 'Barbatimão'). Silica core was synthesized by the modified Stöber method. The pH responsive polysaccharides used to coat the porous silica cores were chitosan, and sodium alginate, this coating was carried out using the Layer-by-Layer technique. The hybrid nanoparticles were characterized at molecular and physical-chemical levels. Furthermore, encapsulation efficiency, pH-dependent release behavior, and cytotoxicity were evaluated. Amorphous mesoporous structure with adequate size for follicular delivery (between 300 and 600 nm) in addition to effective phytocompound loading capacity, above 80 % was obtained. Based on the release studies, it was possible to observe pH responsiveness. The ethyl acetate fraction (EAF) obtained from "Barbatimão" bark extract was released in a controlled and more efficient manner by the alginate-coated nanoparticle (SNP_EAF_SA) at pH 7.4, which corresponds to the pH at the deepest area of hair follicles. Furthermore, SNP_EAF_SA proved to be less cytotoxic compared to EAF and chitosan-coated hybrid nanoparticles (SNP_EAF_CH). Characterization, release, and cytotoxicity results indicate that SNP_EAF_SA is a promising system for on-demand follicular delivery of antiandrogenic actives contained in EAF.

This in vitro study aimed to evaluate the growth-inhibitory effects against periodontal disease-causing bacteria and cytotoxic effects against mouse fibroblast cells of the Stryphnodendron adstringens (barbatimão) hydroalcoholic extract. The contents of phenols and tannins in the extract were determined. The growth-inhibitory activity of the barbatimão was evaluated by determining the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The viability of fibroblast cells was analyzed using the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide assay at 24 and 48 h post-treatment. The MIC values of the extract against Prevotella intermedia, Porphyromonas gingivalis, and Fusobacterium nucleatum were 0.05, 0.125, and 2 mg mL-1, respectively, while the MBC values were 4, 2, and 2 mg mL-1, respectively. The viability rate of barbatimão (0.25 mg mL-1)-treated L929 cells was higher than that of chlorhexidine (0.12%)-treated L929 cells at 48 h post-treatment. The contents of total phenolics and total tannins in the extract were 837.39 ± 0.10 and 785.82 ± 0.14 mg of tannic acid equivalent per gram of the extract, respectively. These findings indicate that the barbatimão hydroalcoholic extract, which exerted potent growth-inhibitory effects against the test microbial species and low cytotoxic effects on fibroblasts, has potential applications in the development of novel mouthwash products.

The present work aimed to evaluate the healing effect of hydrophilic polymeric resorbable biomembrane scaffolds containing plant extracts obtained from two different species, both popularly known as Stryphnodendron adstringens or Barbatimão. The hydrogel-based scaffolds were characterized and submitted to biological tests using Wistar rats to evaluate their healing capacity. The wound retraction index and the evaluation of the inflammatory process and tissue collagenization were recorded. The extracts showed antioxidant activity with IC50 between 10 and 20 µg/mL (DPPH assay) and 4-6 mmol Trolox/g (FRAP assay). The extract of Stryphnodendron adstringens (SA) presented gallocatechin, epigallocatechin, and O-methylpigalocatechin, while the extract of Abarema cochliacarpa (AC) presented catechin, dimers of procyanidins, di-O-hydroxide, O-deoxyhexosi-hexoside, and epicatechin. The membranes containing SA extract (GELSA) were more rigid, with a more intense color, but less thick, with a more compact structure and few pores. The membranes containing AC extract (GELAC) presented a mechanical profile like the gelatin membrane (GEL), with greater permeability to water vapor. The GELAC and GELSA membranes showed similar thermal degradation profiles. The wounds treated with the membranes containing the extracts obtained high levels of retraction of the wounds with values around 60% and 80% in three and seven days, respectively. These data indicate that the compounds of both species have promising biological activities in the repair process, showing that the extracts accelerated the healing process due to the lower intensity of the inflammatory reaction and the presence of compounds such as catechin and epigallocatechin.

Stryphnodendron adstringens is a medicinal plant that has a broad spectrum of action, including antibacterial activity. The aim of the present study was to evaluate the effect of S. adstringens alone and in combination with potassium sorbate (PS) against foodborne bacteria. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined and, for most of the bacteria tested, the crude extract (CE), aqueous fraction (AQF), and ethyl-acetate fraction (EAF) of S. adstringens had a MIC and MBC ranging from 500 to ≥ 1000 µg/mL. The AQF and EAF showed greater activity against S. aureus strains (MIC = 125 to 250 µg/mL; MBC = 500 to 1000 µg/m). Quantitative cell viability was determined and was observed reductions ranging from 3.0 to 5.8 log10 CFU/ml.The combination of S. adstringens and PS against seven S. aureus isolates was determined by the checkerboard method at neutral and acid pH. In a neutral medium, the AQF + PS combination presented synergistic or additive interactions against six S. aureus strains. The combination of EAF + PS resulted in additive interactions against four bacterial isolates. In an acidic medium, the AQF + PS combination was synergistic or additive against all S. aureus, while EAF + PS presented the same effect against six S. aureus strains S. adstringens showed important antibacterial effects against foodborne S. aureus strains. Moreover, the combination of S. adstringens fractions and PS improved the antibacterial activity compared to the compounds utilized individually. The combined use of these compounds may be an alternative to reduce bacterial food contamination and improve food safety.

Breast cancer appears as the main gynecological cancer and presents high morbidity and mortality. Because most diagnoses are made belatedly, it is necessary to seek therapeutic options that aim for advanced stages of the disease. This study aims to evaluate the antitumoral action of Stryphnodendron adstringens fruit extracts on 4T1 murine mammary carcinoma cell culture. The inhibitory potential of S. adstringens fruit extract on the metalloproteinases (MMPs) 2 and 9 was evaluated through zymography. From these results, MTT assays were performed to evaluate the extracts' effects on the murine mammary carcinoma 4T1 line cell viability. From the crude extract, the following extracts were obtained: hydroalcoholic (SAFCEA), hexane (SAFCEB), chloroform (SAFCEC), and ethyl acetate (SAFCED). Lastly, the migration of the cells treated with extracts SAFCEA and SAFCED was verified. The hydroalcoholic extract (SAFCEA) was the most efficient in inhibiting gelatinases. During the phytochemical study, it was noted that alkaloids were present in all partitions. The 50 % growth inhibition (IC50) concentrations found were: 40.1 µg/mL (SAFCEA) and 70.14 µg/mL (SAFCED). After the cellular cytotoxicity assay, cell morphology was altered by treatment with the selected partitions (SAFCEA and SAFCED), obtaining morphology consistent with apoptosis. The results demonstrate that S. adstringens extracts exhibit the inhibitory activity of MMP-2 and MMP-9 as well as cytotoxicity toward 4T1 tumor cells. These findings indicate that follow-up studies of the partitions from S. adstringens may lead to the development of novel chemotherapeutics for oncological treatments.

Stryphnodendron adstringens is a typical tree from Brazilian Savanah used in medicine as an anti-inflammatory and antiseptic agent. It is secondary metabolites has biological activities, so the development of efficient extraction methods is essential. Microwave irradiation through assisted extraction is innovative and highly efficient for bioactive compounds. The aim of this study was to optimize an extractive method for phenolics compounds, as tannins, from the stem bark of "barbatimão" by microwave irradiation using a statistical planning and to evaluate its consistency with conventional extraction. Microwave irradiation extraction, 16.36-22.12% of phenols and 15.91-18.69% of tannins, has a better yield when compared to conventional extraction, 14.99% of phenols and 16.70% of tannins. The method by microwave irradiation is consistent with the conventional one. However, extraction by microwave irradiation had a reduction in reaction time, reagent volume, samples amount and energy consumption when compared to conventional extraction.

Snakebites produce several toxic effects in victims, such as hemorrhage, tissue necrosis, hemostatic, renal, or cardiotoxic alterations, inflammation, and death. To counteract these symptoms, antivenom is the official treatment. Although such therapy prevents death, it does not efficiently neutralize necrosis or other local effects, leading to amputation or morbidities of the affected limb. Therefore, the search for better and more efficient therapies deserves attention; further, plants have been used to ameliorate a number of diseases and medical conditions, including snakebites, for many years. Thus, the aim of this work was to evaluate the antivenom effect of the crude extract, fractions (aqueous and diethyl acetate), and subfractions derived from the aqueous fraction (P1, P2, P3, and P4) of the plant Stryphnodendron adstringens against in vitro (coagulation and proteolytic) and in vivo (edema, hemorrhage, and myotoxic) activities caused by Bothrops jararacussu venom. Overall, all extracts inhibited the toxic effect of B. jararacussu venom, but with different potencies, regardless of whether plant samples were incubated together with venom or injected before or after venom injection into animals; the crude extract and aqueous fraction were found to be the most effective. Indeed, phytochemical and mass spectrometry analysis of S. adstringens samples revealed the presence of flavonols, tannins, and saponins. In conclusion, the plant S. adstringens may represent a promising natural source of molecules to treat the toxic effects associated with envenomation by B. jararacussu snakebites.

We evaluated the activity of the aqueous fraction and the ethyl acetate fraction of Stryphnodendron adstringens against Staphylococcus aureus and proposed their mechanism of action. The antibacterial activity of S. adstringens fractions was evaluated against S. aureus and the cell targets were rated by docking. The fractions showed moderate antibacterial activity against S. aureus without toxicity on two mammalian cell lines. They also showed synergistic antibacterial activity with tannic acid (TA). In silico assays indicated FabG, FabZ and FabI as probable targets. The metabolic pathway for fatty acid biosynthesis in S. aureus was affected by components of S. adstringens. The synergistic effect when combining TA with S. adstringens fractions suggests a natural alternative to S. aureus control. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study describing the possible targets of action of Stryphnodendron adstringens on Staphylococcus aureus. Molecular dynamics simulations showed that the components of S. adstringens affected the metabolic pathway for fatty acid biosynthesis (FAS II) in S. aureus, inhibiting the FabI, FabG and FabZ enzymes. As tannic acid (TA) is a known inhibitor of some targets identified, we showed synergistic antibacterial activity of S. adstringens in combination with TA. This combination did not show toxicity against HaCaT and Vero cells and based on all these results we suggest that S. adstringens can be a natural and sustainable alternative to S. aureus control.

ETHNOPHARMACOLOGICAL RELEVANCE: Stryphnodendron adstringens has been used by indigenous Brazilian people to treat wound, infections, inflammation and other conditions. AIM OF THE STUDY: This study aims to investigate the effect of S. adstringens on macrophage polarization. METHODS: To prepare the hydroethanolic extract of Stryphnodendron adstringens (HESA), fresh bark was exposed to maceration, filtered and subsequently lyophilized. The extract HESA were analyzed by LC-DAD-MS to identify their constituents. Bone marrow cells were obtained from male C57BL/6 mice. Then, the cells were polarized into M1 or M2 subsets in the presence or absence of HESA. The membrane expression of TLR2, CD206, CCR7, class II MHC, and CD86, the intracellular expression of iNOS and IL-6 and the supernatant expression of IL-6 were determined by flow cytometry. RESULTS: By LC-DAD-MS, twenty-four compounds could be detected from HESA and proanthocyanidins, flavan-3-ols, and chromones were identified. NO and iNOS were reduced in the HESA-treated cells. There was a reduction in IL6 in HESA-treated cells. The membrane expression of TLR2, CD206, CCR7, CD86, and class II MHC was reduced in HESA-treated cells. The densities of CD206 and IL-10 were found to be significantly increased in HESA-treated cells. CONCLUSION: This work is the first to demonstrate that S. adstringens can modulate the functional polarization of macrophages into the M2 profile and suppress costimulatory molecules in M1 macrophages. These results corroborate with the ethnopharmacology use of S. adstringens, contributing to its pharmacological validation in wound treatment and expanding the knowledge about immunoregulatory action of this specie.

ABSTRACT Background: There is still a need for progress in the treatment of transsphincteric anal fistulae and the use of herbal medicines seems promising. Aim: To evaluate the efficacy of Stryphnodendron adstringens associated with fistulotomy and primary sphincteroplasty in the treatment of transsphincteric fistulae in rats. Methods: Thirty Wistar rats were used, which were submitted to transsphincteric fistulas with steel wire 0; after 30 days a treatment was performed according to the group. Group A (n=10) was submitted to fistulotomy; group B (n=10), fistulotomy followed by primary sphincteroplasty with "U" stitch with polyglactin 911 4-0; group C(n=10) , similar to group B, but with the interposition between the muscular stumps of hemostatic sponge soaked in Stryphnodendron adstringens extract. Euthanasia was performed after 14 days, resecting a segment of the anal canal for histological analysis, which aimed to evaluate the closure of the fistula, the area of separation of the muscle cables, the inflammatory process and the degree of fibrosis. Results: No animal had a remaining fistulous tract. About the spacing between the muscle cables, an average of 106.3 µm2 was observed in group A, 82.8 µm2 in group B and 51.8 µm2 in group C (p<0.05). There was no difference between the groups regarding the inflammatory process and, in relation to fibrosis, in group A there was a mean of 0.6, in group B 0.7 and in group C 0.2 (p<0.05). Conclusions: Stryphnodendron adstringens extract was able to allow less spacing between muscle cables in rats submitted to fistulotomy followed by primary sphincteroplasty, in addition to providing less local fibrosis.


RESUMO Racional: Há ainda necessidade de avanço no tratamento das fístulas transesfincterianas e o uso de fitoterápicos parece promissor. Objetivo: Avaliar a eficácia da Stryphnodendron adstringens associada à fistulotomia e esfincteroplastia primária no tratamento de fístulas transesfincterianas em ratos. Métodos: Utilizou-se 30 ratos Wistar submetidos à confecção de fístulas transesfincterianas com fio de aço 0; após 30 dias realizou-se tratamento de acordo com o grupo. O grupo A (n=10) foi submetido à fistulotomia; o grupo B (n=10) fistulotomia seguida de esfincteroplastia primária com ponto em "U" com poliglactina 911 4-0; o grupo C (n=10), semelhante ao grupo B, porém com a interposição entre os cotos musculares de esponja hemostática embebida em extrato de Stryphnodendron adstringens. Realizou-se eutanásia após 14 dias, ressecando-se segmento do canal anal para análise histológica, que objetivou avaliar o fechamento da fístula, a área de afastamento dos cabos musculares, o processo inflamatório e o grau de fibrose. Resultados: Nenhum animal apresentou trajeto fistuloso remanescente. Quanto ao afastamento entre os cabos musculares observou-se média 106,3 µm2 no grupo A, 82,8 µm2 no grupo B e 51,8 µm2 no grupo C (p<0,05). Não houve diferença entre os grupos quanto ao processo inflamatório e, em relação à fibrose; no grupo A observou-se média 0,6, no grupo B 0,7 e no grupo C 0,2 (p<0,05). Conclusões: O extrato de Stryphnodendron adstringens foi capaz de permitir menor afastamento entre os cabos musculares em ratos submetidos à fistulotomia seguida por esfincteroplastia primária, além de proporcionar menor fibrose local.

ABSTRACT: Fasciolosis has been diagnosed in cattle, goats, sheep and horses in southern and southeastern Brazil. Effective alternative treatments are the targets of study. One promising alternative is the use of plant extracts. The aim of this study was to perform phytochemical analysis of extracts of Eugenia uniflora L., Harpagophytum procumbens, Psidium guajava L. and Stryphnodendron adstringens, and to evaluate the in vitro efficacy of these extracts on ovicidal activity in Fasciola hepatica. Plant extracts were analyzed for phytochemical properties. F. hepatica eggs were collected directly from the gallbladders of animals diagnosed as positive for fasciolosis on post mortem examination. One hundred eggs were incubated with 3 ml of each extract at concentrations of 0.10%, 0.25% and 0.50%, albendazole 0.50% (positive control) or tap water (negative control). To determine anti larval efficacy of each plant extract, hatched eggs were counted and the averages were used. Phytochemical analysis revealed the presence of phenolic compounds, tannins and terpenes in most extracts. E. uniflora L. extract was 100% effective at 0.10%, H. procumbens was effective at 0.25% and P. guajava L. and S. adstringens extracts were 100% effective at all concentrations tested. Taken together, the data suggested that ovicidal activity in F. hepatica is due to the presence of these bioactive compounds.


RESUMO: A fasciolose tem sido diagnosticada em bovinos, caprinos, ovinos e equinos no sul e sudeste do Brasil, sendo que tratamentos alternativos mais eficazes são alvos de estudo. Umas das alternativas promissoras é o uso de extratos vegetais no controle dessa e outras enfermidades. O objetivo deste estudo foi realizar a análise fitoquímica dos extratos de Eugenia uniflora L., Harpagophytum procumbens, Psidium guajava L. e Stryphnodendron adstringens, além de avaliar a eficácia in vitro desses extratos na atividade ovicida em Fasciola hepatica. Os extratos vegetais foram obtidos e analisados para determinação fitoquímica. Ovos de F. hepatica foram coletados diretamente das vesículas biliares de animais diagnosticados como positivos para fasciolose no exame post mortem. Cem ovos foram incubados com três mililitros de cada extrato nas concentrações de 0,10%, 0,25% e 0,50%, de albendazol a 0,50% (controle positivo) e água de torneira (controle negativo). Para determinar a eficácia de cada extrato vegetal os ovos eclodidos foram contados, e a média utilizada para os cálculos de eficácia. A análise fitoquímica revelou a presença de compostos fenólicos, taninos e terpenos na maioria dos extratos. O extrato de E. uniflora L. apresentou eficácia de 100% na concentração de 0,10%, o de H. procumbens a 0,25% e os extratos de P. guajava L. e S. adstringens apresentaram 100% de eficácia em todas as concentrações testadas. Assim, sugere-se que a atividade ovicida em F. hepatica seja devido à presença desses compostos bioativos.

Medicinal plants have been recognized as hosts of high diverse endophytic microorganisms, including fungi that produce secondary metabolites with biological activity. Two biomes in Brazil, Pantanal (wetland), and Cerrado (savannah), are known as biodiversity hotspots, and despite their importance as a reservoir for several species, knowledge about the fungal biodiversity in these biomes is very limited. Fungal endophytic communities associated with leaves and petioles of the medicinal plants Vochysia divergens (from Pantanal) and Stryphnodendron adstringens (from Cerrado) were analyzed and studied for their antimicrobial activity against human and plant pathogens. A total of 1,146 isolates of endophytic fungi were obtained from plants collected in January and June of 2016 and grouped into 124 morphotypes. One isolate of each morphotype was identified by sequencing of internal transcribed spacer (ITS) region of the rDNA gene, which revealed the presence of 24 genera, including 3 possible new genera, and 48 taxa. Differences in the endophytic community according to the biomes were observed concerning the analyzed morphotypes. However, when we analyzed the diversity of genera and richness, they were similar for both plants, with Diaporthe, Phyllosticta, and Neofusicoccum as dominant genera. In addition, the community composition of V. divergens differs according to the analyzed plant tissues (petiole and leaf). These data suggested that both, the plant species and plant tissues play a role in the composition of endophytic community. As regards the biotechnological potential, 5 isolates showed activity against the phytopathogens Phyllosticta citricarpa, Colletotrichum abscissum, and Fusarium verticilioides, and 8 isolates showed high activity against clinical pathogens and were selected for the production of crude extract in different culture media. Extract from cultivation of Diaporthe sp. LGMF1548 and LGMF1583 and Neofusicoccum brasiliense LGMF1535 showed activity against methicillin-resistant Staphylococcus aureus, Klebssiella pneumonia, and Candida albicans. In addition, extracts of Diaporthe cf. heveae LGMF1631 inhibited 90% of the mycelial growth of the P. citricarpa and 70% of C. abscissum and may represent an alternative to be used in the biological control of these phytopathogens. Future research will focus on the chemical characterization and structural elucidation of these bioactive compounds.

In vitro bioassay using Sarcoptes scabiei as a test microorganism is a viable method of study for diverse drugs with acaricidal properties. A great amount of assays proves the diverse and efficient biological activity of extracts and compounds from Brazilian savanna plants. This study had as main object, test and compare the acaricidal activity of four Brazilian Cerrado bioproducts: Stryphnodendron adstringens Mart., Copaifera sp., Lafoensia pacari A. St Hil. and Pterodon emarginatus Vogel, 1837. To perform this test S. scabiei mites were collected from crusts removed on ears of highly infected sows. The mites were selected and separated in Petri dishes with three different concentrations (25, 50 and 75%) of each bioproduct, to evaluate their potential acaricidal activity. The mortality of the mites was counted in each Petri dish every hour, during five hours. The statistical analyses demonstrated differences between the bioproducts tested. The oleoresin of Copaifera sp. and P. emarginatus presented the best results with 100% of mites mortality after treatment. The ethanolic extracts of S. adstringens and L. pacari demonstrated lower acaricidal activity when compared to the oleoresins, with little or no difference among the control groups tested. This bioassay demonstrated to be efficient, reliable, low cost and easy accomplishment. Oil resins from Copaifera sp. and P. emarginatus have in vitro acaricidal activity against adult females of S. scabiei var. suis.(AU)


O uso de Sarcoptes scabiei como microrganismo teste para bioensaio in vitro é um método de estudo viável para diversas drogas acaricidas. Muitos ensaios comprovam a diversidade e eficiência de atividade biocida de extratos e componentes presentes em plantas do cerrado brasileiro. Este trabalho objetivou testar e comparar a atividade acaricida de quatro bioprodutos do cerrado brasileiro: Stryphnodendron adstringens Mart., Lafoensia pacari A. St Hil., Copaifera sp. e Pterodon emarginatus Vogel, 1837. Para este estudo, ácaros S. scabiei foram colhidos em crostas removidas de orelhas de fêmeas suínas altamente infestadas. Os ácaros foram selecionados, separados e colocados em placas de Petri com três diferentes concentrações (25, 50 e 75%) de bioprodutos, para avaliar o potencial de atividade acaricida de cada um. Foi realizada a contagem da mortalidade dos ácaros em cada placa de Petri a cada hora, durante cinco horas. As análises estatísticas demonstraram diferenças entre os bioprodutos testados. As óleo-resinas de Copaifera sp. e P. emarginatus apresentaram os melhores resultados, com 100% de mortalidade dos ácaros após tratamento. Os extratos etanólicos de S. adstringens Mart. e L. pacari demonstraram menor atividade acaricida quando comparados as óleo-resinas, com pequena ou nenhuma diferença entre os resultados dos grupos controle. Este ensaio demonstrou ser uma ferramenta eficiente, confiável, de baixo custo e de fácil realização. As óleo-resinas Copaifera sp. e P. emarginatus possuem atividade acaricida in vitro sobre fêmeas adultas de S. scabiei var. suis.(AU)

Objetivou-se com este estudo avaliar o processo de cicatrização de feridas de coelhos tratadas com extrato de barbatimão (S. adstringens) associado a células mononucleares autólogas da medula óssea (CMMO). Utilizaram-se 20 coelhos, distribuídos em quatro grupos: B, extrato de barbatimão; CB, CMMO com extrato de barbatimão; CS, CMMO com solução fisiológica; S, solução fisiológica. Foi avaliada a presença de crosta, hiperemia, secreção, hemorragia, reepitelização, área da ferida e tempo de cicatrização. No terceiro, sétimo, 14º e 21º dias pós-operatório, realizou-se a biópsia das feridas e avaliaram-se os indicadores dos processos de inflamação e de reparo, com destaque para o colágeno, na coloração picrosírius, bem como de proliferação celular, na coloração AgNOR. Houve maior deposição de fibras colágenas nos grupos B e CB (P=0,00003) e formação de crostas mais espessas no sétimo dia, com fibras colágenas mais organizadas no 21º dia. Conclui-se que o barbatimão estimula a produção de fibras colágenas e promove a formação de crostas mais espessas sobre a ferida na fase inicial da cicatrização e, na fase de remodelação, favorece a orientação das fibras colágenas. Além disso, a associação desse fitoterápico com CMMO não estimula a cicatrização de feridas.(AU)


This study aimed to evaluate the healing process of wounds of rabbits in response to treatment with barbatiman extract (S. adstringens) associated with autologous mononuclear bone marrow cells (BM-MNC). We used 20 rabbits, divided into four groups: B, 10% barbatiman extract with 9.48% of total tannins; CB, BM-MNC with barbatiman extract; CS, BM-MNC with NaCl 0.9% solution; S, NaCl 0.9% solution. Clinical evaluation was performed by observing the presence of crust, redness, discharge, bleeding, re-epithelialization, the wound area and healing time in days. In the third, seventh, 14th and 21st postoperative days wounds were biopsied for microscopic evaluation of inflammation and repair process indicators, especially collagen, in picrosirius staining, and cell proliferation, in AgNOR staining. There was a greater deposition of collagen fibers in groups B and CB (p=0.00003) on the seventh day and formation of thicker crusts, and more organized collagen fibers on the 21st day in these groups. In conclusion, in the initial phase of healing, barbatiman extract stimulates the production of collagen fibers and promotes the formation of more exuberant crusts on the wounds and remodeling phase favors the orientation of collagen fibers, but when combined with BMMC does not stimulate wound healing in rabbits.(AU)

This study evaluated the antimicrobial activity and acute or chronic toxicity of the extract of Stryphnodendron adstringens. The stem bark dry extract was obtained by static maceration with ethanol. Quantification of tannins was performed by the Folin-Denis method, which indicated a total tannin content of 32.7%. The antimicrobial activity of the dry extract of S. adstringens was evaluated by agar-based disk diffusion assay with Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923) in the concentration of 200, 400 and 600µL/mL. The results indicated that 600µL/mL inhibited microbial growth, i.e. had antimicrobial activity against these species. Acute and chronic toxic effects of S. adstringens was evaluated in Wistar rats treated with 200, 400, 600 and 800mg/kg of extract, administrated by gavage. Liver degeneration was observed in the group of rats receiving 800mg/kg in chronic exposure, what may indicate some degree of toxicity at this concentration. However, no systemic toxicity was observed at lower doses. Considering the broad use of S. adstringens as a phytotherapeutic agent for various human and animal diseases and the livertoxicity observed at high concentrations, attention should be paid to the possible adverse effect of using the extract from this plant at high concentration.(AU)


Avaliou-se neste estudo a atividade antimicrobiana e a toxicidades aguda e crônica do extrato da entrecasca de Stryphnodendron adstringens. A partir do extrato seco, obtido através da maceração estática da casca do caule em etanol de cereais, foi realizada a quantificação de taninos totais (32,7%) pelo método de Folin-Denis. A atividade antimicrobiana do extrato seco extraído de cascas do caule de S. adstringens foi avaliada pela técnica de disco-difusão para os micro-organismos Escherichia coli (ATCC 25922) e Staphylococcus aureus (ATCC 25923) nas concentrações de 200, 400 e 600µL/mL. Os testes de concentração inibitória mínima revelaram que 600µL/mL inibiu o crescimento dos dois micro-organismos testados; o mesmo resultado foi observado para atividade bactericida na concentração de 600µL/mL sobre essas espécies. Efeitos tóxicos sistêmicos agudos e crônicos do extrato seco de S. adstringens administrados por gavagem foram estudados em ratos Wistar, utilizando as doses de 400, 600 e 800mg/kg. Foi observada degeneração hepática no grupo de animais que receberam 800mg/kg tanto no estudo da toxicidade aguda quanto crônica, que pode indicar algum grau de toxicidade de S. adstringens nessa concentração. Considerando o amplo uso de S. adstringens como fitoterápico para humanos e animais, atenção deve ser dispensada para ingestão em altas doses mediante os efeitos tóxicos observados neste estudo.(AU)

This study evaluated the aqueous extraction of galactomannans from the seeds of Mimosa scabrella (GM), Stryphnodendron adstringens (GS) and Schizolobium parahybae (GG) for 1, 2, 3, 4, 6, 24 and 48 h. The efficiency of extraction processes was assessed in terms of yield, carbohydrate and protein content. The extraction process, as well as the source of the galactomananns generated molecules with differences in molar mass, viscosity and rigidity analyzed by HPSEC-MALLS/RI/VIS. The extraction time results for each species, based on minimum extraction time and HPSEC-MALLS/RI/VIS results, were 4 h (GM4h), 6 h (GS6h) and 2 h (GG2h) for GM, GS and GG, respectively. In most cases, the apparent persistence length, as determined by viscometry, indicated that aggregates remained in galactomannans after centrifugation and filtration. Results suggest an effective extraction time for each plant source of galactomannan based on its performance and its macromolecular behavior in solution.

BACKGROUND: Stryphnodendron adstringens (Mar.) Coville is a native plant from Brazil, rich in phenolic compounds and used on popular medicine as a wound healing agent, in the treatment of gastric lesions and as antimicrobial. MATERIALS AND METHODS: Ultrassound-assisted extraction (UAE) was applied to extraction of epigallocatechin gallate (EGCG), total polyphenols (TP) and total tannins (TT) content from barks of Stryphnodendron adstringens (Mar.) Coville. Several operating parameters, namely extraction time (min), liquid to solid ratio (mg/mL), ethanolic strength (%, v/v), were optimized using response surface methodology (RSM) with a Box-Behnken design. RESULTS: By using the desirability function approach, the optimum UAE conditions to obtain desirable extraction yields for all these metabolites simultaneously were found at the extraction time of 30 min, solid to liquid ratio of 4 mg/mL and ethanolic strength of 65. Under these conditions, the epigallocatechin gallate, total polyphenols and total tannins content were 0.31; 22.95 and 11.95 % (w/w), respectively. CONCLUSION: The results indicated that knowledge gained from this study should be helpful to further exploit and apply this resource and also showed the feasibility of ultrasound-assisted extraction for obtaining GEGC, TP and TT from barks of S. adstrigens.

The aim of this study was to evaluate the insecticidal effect of aqueous, alcoholic, and oil extracts from leaves of eight medicinal plants against Diabrotica speciosa prepared at five concentrations. The extracts that used commercial soybean oil as solvent showed the highest D. speciosa mortality due to the solvent itself, regardless of the used plants and their concentrations. Thus, commercial soybean oil was discarded as solvent since at these volumes it would cause serious phytotoxicity problems. After 24 hours of exposure of the pest to the extracts, the highest D. speciosa mortality values were observed for Copaifera langsdorfii and Chenopodium ambrosioides extracts, both in 5% alcohol, and Artemisia verlotorum, in 10% water. However, in the last mortality assessment (48 h), C. langsdorfii extract in 5% alcohol showed higher mortality of this pest, followed by C. ambrosioides extract in 5% alcohol, compared to the remaining plants.


O objetivo deste trabalho foi avaliar o efeito inseticida de extratos aquosos, alcoólicos e oleosos de folhas de oito plantas medicinais contra Diabrotica speciosa preparadas em cinco concentrações. Os extratos que utilizaram óleo de soja comercial como solvente apresentaram as maiores mortalidades de D. speciosa em função do próprio óleo, independentemente das plantas utilizadas em suas concentrações. Sendo assim, o óleo de soja comercial foi descartado como solvente, pois nestes volumes acarretaria sérios problemas de fitotoxidade. Após 24 horas de exposição da praga aos extratos, os maiores valores de mortalidade de D. speciosa foram observados nos extratos de Copaifera Langsdorfii e de Chenopodium ambrosioides, ambos em álcool 5%, e de Artemisia verlotorum, em água 10%. Entretanto, na última avaliação de mortalidade (48 h), o extrato de C. langsdorfii em álcool a 5% apresentou maior mortalidade dessa praga, seguida pelo extrato alcoólico a 5% de C. ambrosioides comparada às demais plantas.

Stryphnodendron adstringens (Mart.) Coville (barbatimão) is a tree belonging to the Fabaceae family, and it is commonly found in the southeastern Brazilian cerrado. The stem bark of this tree is traditionally used as an anti-inflammatory and antiseptic agent to treat leucorrhea and diarrhea, as well as to promote wound healing (owing to the presence of proanthocyanidins). Proanthocyanidins were obtained from the ethanolic extract of S. adstringens stem bark and assessed by reversed phase-high performance liquid chromatography with an ultraviolet/diode array detector. The identified compounds included gallic acid, catechin, gallocatechin (GC), epigallocatechin, and epigallocatechin gallate (EGCG). The selected markers, GC and EGCG, were simultaneously used for chromatographic validation (linearity range: 30-330 ng, equivalent to 3-33 µg/mL; r>0.998). The method showed precision (intra-day relative standard deviation [RSD]: 1.72% for GC; 1.16% for EGCG; inter-day RSD: 1.74%-2.60% for both markers), accuracy, robustness, and selectivity. The limits of detection and quantitation were 0.29 µg/ml and 0.89 µg/ml for GC, and 0.88 µg/mL and 2.67 µg/mL for EGCG, respectively. In addition, S. obovatum was evaluated and showed an average of 12.2 µg/mL for GC (equivalent to 1.22% w/w) and 14.2 µg/mL for EGCG (equivalent to 1.42% w/w) in the ethanolic extract. The quantitative results were compared to those obtained for S. adstringens, which showed that the markers are present in both species.


Stryphnodendron adstringens (Mart.) Coville (barbatimão) é uma árvore da família Fabaceae encontrada no cerrado do sudeste do Brasil. As cascas do caule das espécies são tradicionalmente utilizadas como agente antiinflamatório e antisséptico para o tratamento de diarreia e leucorreia, bem como para promover a cicatrização de feridas, devido à presença de proantocianidinas. Estas substâncias obtidas a partir do extracto etanólico (EE) de cascas do caule de S. adstringens foram avaliadas por cromatografia líquida de alta eficiência de fase reversa. As substâncias identificadas foram ácido gálico, catequina, galocatequina (GC), epigalocatequina e galato de epigalocatequina (EGCG). Os marcadores selecionados, GC e EGCG, foram utilizados simultaneamente para validação cromatográfica (intervalo de linearidade 30-330 ng, equivalente a 3-33 µg/mL, r>0,998). O método mostrou precisão (intra-dia DPR 1,72% para GC; 1,16% para EGCG; inter-dia DPR 1,74%-2,60% para ambos os marcadores), exatidão, robustez e seletividade. Os limites de detecção e de quantificação de 0,29 µg/mL e 0,89 µg/mL para a GC, e 0,88 ng/mL e 2,67 µg/mL para EGCG, respectivamente. Outra espécie (S. obovatum) avaliada, mostrou valores médios 12,2 µg/mL de GC (equivalente a 1,22% p/p) e 14,2 µg/mL de EGCG (equivalente a 1,42% p/p) encontrados no EE. Os resultados quantitativos comparam-se aos de S. adstringens e mostram que tais marcadores estão presentes em ambas as espécies.
